Thallium Flux
Concentration-dependent effects of positive (PAM) and negative (NAM) allosteric modulators of the G-protein-gated inward rectifying potassium channel (GIRK). Fluorescence measurement of thallium (Tl+) flux were obtained on PANOPTIC using the Tl+-sensitive dye, Thallos, at 1 Hz (480/40x, 535/40m).
Concentration-dependent effects of veratridine (an inhibitor of voltage-gate sodium channel inactivation) on NaV1.7-expressing HEK-293 cells. Fluorescence measurement of thallium (Tl+) flux were obtained on PANOPTIC using the Tl+-sensitive dye, Thallos, at 5 Hz (480/40x, 535/40m). Veratridine concentrations 10 uM – 10 nM in 3-fold steps.
Luminescence
Concentration-dependent effects of a-MSH (an agonist of the Gs-coupled GPCR, MC4R) on HEK-293 cells expressing MC4R and Promega’s GloSensorTM cAMP-sensitive luciferase. Luminescence measurement of the agonist-evoked increase in intracellular cAMP were obtained on PANOPTIC at 1/3 Hz with a 720 nm short pass emission filter, 37 C. Calcium Flux
Concentration-dependent, ATP-stimulated increases in cytoplasmic and mitochondrial Ca2+ in HEK-293 cells loaded with Fluo-8 and Rhod-2. Dual excitation, dual emission data were obtained on PANOPTIC at 1 Hz using filter sets appropriate for Fluo-8 (470/40x, 525/30m) and Rhod-2 (545/30x, 593/40m).
Neurotransmitter Transport
Concentration-dependent effects of Desipramine on HEK-293 cells expressing the norepinephrine transporter (NET) using the Molecular Devices Neurotransmitter Transporter Uptake Assay Kit.
